A tender (small boat) from Santa Regina picked up one person in the water who was then transferred to a Coastguard boat and taken to Blenheim by ambulance.
The body of a second person was recovered by a local water taxi and taken to shore.
The people in the water are believed to have been in a 10-metre runabout but details of the incident are yet to be confirmed.
Santa Regina has returned to Picton.The Maritime Safety Authority will launch an immediate investigation.
